Hearts of Oak have been paired with Liberty Professionals while Asante Kotoko will face Berkwai Youth FC in the MTN FA Cup round 64 after the draw was held on Thursday.
Hearts of Oak who have won the competition a record 10 times must be at their best to overcome their regional rivals and reach round of 32.
Kotoko, on the other hand, are clear favourites against the second-tier side. Kotoko and Berkwai met at the same stage in 2017 with the Porcupine Warriors winning on penalties after a 1-1 stalemate in regulation time. Kotoko went on to win the tournament, beating Hearts of Oak 2-1 in the final in Tamale.
Other exciting fixtures include Aboi United FC versus Samartex 1996.
Two time winners Medeama SC will face Sekondi Eleven Wise – with 1985 champions Okwaku United set to face Division Two side Kade United FC.
Matches in the Round of 64 are scheduled for Friday, May 28 to Monday, May 31, 2021.
